Abstract

Superior upland rice cultivar can be assembled through crossbreeding using germplasm diversity which serves as a sourceof gene. Germplasm may constitute of landrace and local cultivars. Banten is one of the provinces that has various localupland rice varieties. Baduy tribe, in the province has played a major role in preserving local upland rice varieties anduphold the wisdom.Genetic diversity information and the benefits of local cultivars can be useful to maximize the selectionprocess of parental candidate for creating new cultivars. This study aims to determine the genetic diversity of local uplandrice from Banten. Methods of analysis diversity performed by phenotypic and genotypic. Observations phenotypic throughobservation of agronomic characters and yield of each cultivar. Whereas genotypic observations conducted with RAPD(random amplified polymorphism DNA). In this study we used 17 local upland rice cultivars from Banten with 14 randomprimers for RAPD. The result showed a mean plant height local upland rice cultivar of Banten was higher than nationalupland rice varieties however productive tillers and rice productivity were lower than national varieties. Nonetheless, localcultivars persisted in this environment because it has the advantage that the community needed. Based on the time ofharvest, local cultivar belonged to early and medium maturing group. Qualitative trait observation showed that there werediversity of stem, leaves, grain and rice among the cultivars. Genetic analysis with RAPD showed nine primers out of 14were successful in amplifying loci from DNA template. The primers having high amplification product were OPJ 05, OPJ07 and OPD07 but the highest polymorphism presentation was OPJ 01. Based on phylogenetic tree, there were two mainclusters. Padi Koneng and Carogol were similar. Three glutinous rice cultivars (Mayang, Jalupang, and Hideung) were inone cluster except Kentan Simpai. Keywords: upland rice, Banten, RAPD, Phenotipe, genotype

Abstract

Patients with H5N1 disease have a characteristic complications of pneumonia with acute respiratory syndromedisterss and multi-organ failure associated with cytokine dysregulation, so it is suspected that the clinical forms ofsevere H5N1 disease in humans is caused by dysregulation of cytokine induction of H5N1 virus. Study oncytokines associated with H5N1 influenza virus infection is still limited, while animal studies using virus H5N1/97give conflicting results. Recent studies, showing that in fact the high cytokine is not a common form of the H5N1viruses, so Pathogenecity of H5N1 viruses were not only determined by whether or not the virus can inducehypercytokinemia. In this study we wanted to determine the phenotype of the Indonesia H5N1 virus in inducingcytokine expression. This study compared three Indonesian H5N1 viruses that have different genetic variations ofthe virus including the clade 2.1.1 and two other viruses included in clade 2.1.3 that were infected in Madin DarbyCanine Kidney (MDCK) cell. The MDCK cell that infected virus were observed for 72 hours. We observed theinfection during 72 hours and the cytokines expression were examined using RT-PCR method. The results showedthat the H5N1 influenza viruses used in this study showed the low expression of cytokines IFN-γ, and did notshow any expression of the TNF-α cytokines in MDCK cells infected with the H5N1 virus within 72 hours afterinfection. Keywords : Expression, H5N1 virus, Cytokines, sel Madin Darby Canine Kidney (MDCK)

Abstract

Isolation of antibacterial compounds from chloroform extract of belawan putih (Tristaniopsis whiteana) leaves hasbeen done. The bioautography analysis of the extract against Escherichia coli and Staphylococcus epidermidis, subsequently followed by isolation and purification of active compounds led us to obtain two pure compounds, compound 1 and 2. Futhermore, the compound 2 was identified as betulinic acid based on their spectroscopic dataincluding ultraviolet spectrum, infrared spectrum, 1D- and 2D-NMR spectra. In addition, this is the first report ofthe occurrence of this compound in the plant of Tristaniopsis whiteana and the genus of Tristaniopsis. Keywords: antibacterial, active compound, leaves, Tristaniopsis whiteana

Abstract

The animal obtains energy from foods. Apparent Metabolizable Energy (AME) is used to determine metabolizableenergy value of bird’s food. The objective of this research was to evaluate Apparent Metabolizable Energy (AME)value in Blue-crowned hanging parrot (Loriculus galgulus L., 1758), fed with corn mashes, oat mashes, dan pollardmashes. This research consisted of two experiments, which were 1 week of preliminary study and 8 weeks of datacollection. Studies were carried out with six Blue-crowned hanging parrots, two males and four females. The birdswere kept individually in metabolism cage (70 x 43 x 52 cm). The birds were treated with 3 different energy sources,corn-based diet (P1), oat-based diet (P2), and pollard-based diet (P3). The diets were then offered to the birds adlibitum in the form of mashes. Variables observed include dry matter consumption and Apparent MetabolizableEnergy (AME) value. From the calculation, AME of birds that fed with pollard mashes was higher compared to cornand oat mashes as well. In P1, AME value of birds was 43.64 cal/g with 93.90% of metabolic efficiency. While, inthe P2 and P3, AME value were 43.86 cal/g and 39.07 cal/g with number of metabolic efficiency 91.75% and94,58% respectively. Keywords: nutrition, pollard, oat, corn, AME, metabolic efficiency

Abstract

Atactodea striata (Gmelin, 1791) are inhabitants of the sandy at Village Ohoider, Kei Kecil Islands, MalukuTenggara. Sampling has been carried out on the 15th of September 1997 to October 1998 for the shell heightmeasurement. Monthly length frequency data were then analyzed using the software FiSAT II to estimatepopulation parameters such as population structure, age maximum, maximum length, growth and recruitment.Result obtained from this study is that Atactodea striata shells generally consist of four age groups, except in certainmonths were widened to five because of the occurence of old shell group. Mussels in this study reproducemultivoltine with expected life span of almost four years. The maximum length of the mussels is 19.03 mm at 95%confidence level, and the growth coefficient K = 0.37 with a growth performance index φ = 1.74. Observations onhabitat sediment discovered that these shells prefer to live in the fine sand with range of fire grain of 65.96 to69.09%.Keywords:: Dynamics Population, Atactodea striata, Kei Kecil Islands, Maluku Tenggara

Abstract

Gembili is a tuber plant which is usually used as an alternative food source. It containts high carbohydrates andpotentially to be developed into flour. Fermentation of gembili is the method to improve the quality of gembiliflour. The objective of this research was to study the physicochemical properties of fermented gembili flour with theaddition of cellulolytic (BS) and lactic acid bacteria (BAL). There were four treatments, namely naturalfermentation without the addition of neither BS nor BAL, with the addition of BS, with the addition of BAL, andwith the addition of BS and BAL. Fermentation was carried out for three days. Temperature, pH, the number ofbacteria, as well as cellulase and amylase activity were observed every day. The studies of physical properties ofgembili flour were the color, flavor, texture, and yield. Furthermore, the chemical properties included water, ash, carbohydrate, fat, and protein content. Data were analyzed by ANOVA and continued by LSD test at 5%significance level. It showed that the physical properties of flour color in the treatment of BS fermentation werebetter than other treatments. However, the aroma and texture did not show any significant difference. Thechemical properties showed no significant difference among all treatments. Keywords: fermented gembili flour, physicochemical

Abstract

Tree architectural model is basically a tree construction as a result of meristematic growth pattern. Tree architectureis closely associated with water and soil components, i.e. rainfall, throughfall, stem flow, infiltration, surface run-off,and erosion. For Altingia excelsa (Rasamala) plots, the results showed that the daily average of the rainfall intensityobserved was 9.67 mm, stem flow 0.03 mm, canopy throughfall 5.43 mm, infiltration 0.51 ml/mm2/second,surface run-off 3.45 mm, and erosion 5.66 kg/m2. For Schima wallichii (Puspa), the daily average of rainfall was9.67 mm, stem flow 0.04 mm, canopy throughfall 4.02 mm, infiltration 0.49 ml/mm2/second, surface run-off 8.18mm, and erosion 12.71 kg/m2. Compared to A. excelsa, S. wallichii significantly had larger values in someparameters measured, i.e. stem flow, surface run-off, and erosion, indicating that on land sopes of 70% of theMount Gede Pangrango National Park, individual plants of S. wallichii seemed to be well adapted. However, individuals of A. excelsa possessed a more spreadly branching model, larger vertical width, denser canopy, andskewed bark channels capable for slowing down the stem flow and canopy throughfall. Consequently plants of A.excelsa would generally be able to hold and conserve water and soil better than S. wallichii plants. Keywords: Tree architectural model, stem flow, throughfall, surface run-off, infiltration, erosion
